For details including a precision analysis, see document QF2024-020_OXY*.xlsx. Salinity samples were collected in 200 mL or 250 mL type II glass bottles with disposable nylon inserts and screw caps supplied by Ocean Scientific International Limited. They were analyzed in a temperature-controlled lab on a Guildline 8400B Salinometer standardized with IAPSO standard seawater within 27-36 days after collection. For details, see document QF 2024-020 SAL_*.xls. Nutrient samples were collected in plastic tubes and quick frozen in aluminum blocks stored in a -20C freezer. For samples 400m and deeper a second set of samples are collected and stored cool for silicate analysis. All samples were returned to the Institute of Ocean Sciences and they were analyzed using an Astoria-Pacific Analyzer following methods described in IOS Nutrient Methods (1996) Barwell-Clarke and Whitney. Chlorophyll samples were filtered onto 25mm GF/F filters after collection, and immediately frozen in liquid nitrogen and stored in a -80C freezer until analysis. All samples were returned to the Institute of Ocean Sciences and analyzed 1-6 months after collection on a Turner Trilogy7200-000 fluorometer, using methods described in Holm-Hansen et al. (1965). Fluorescence readings taken before and after sample acidification were used to calculate chlorophyll and phaeopigment concentrations. No flags or comments are assigned for Phaeo-Pigment values The Ammonium method used at IOS is a modification of the fluorometric technique of Holmes et al., 1999. The method uses a single working reagent and minimizes contamination, can be applied to a large range of ammonium concentrations, and allows for samples to be processed quickly in the field. Samples are collected during the day into vials cleaned with working reagent, and stored in the Cold Room on the Tully. Working standards that bracket the expected sample concentrations are made up daily from seawater low in organic matter, collected on 2023-066, station P23, at 2005m. Samples and standards are spiked with working reagent at the same time, and allowed to incubate for 8-18 hours to maximize fluorescence signal and stability. The fluorescence is then read using a Turner Trilogy fluorometer in raw fluorescence mode. A linear calibration curve is generated from the standards, and the calibration equation then applied to sample fluorescence to determine sample concentration.
