Salinity samples were collected in 200 mL type ll glass bottles with disposable nylon inserts and screw caps supplied by Ocean Scientific International Limited. They were analyzed in a temperature-controlled lab on a Guildline 8400B Salinometer standardized with IAPSO standard seawater 20-35 days after collection. For details see document QF2023-029_SAL*.xlsx. CHL-a Analysis Details: Seawater samples were collected from niskin bottles and freshly filtered onto 25mm GF/F filters at 5Hg pressure using a vacuum pump at 5"Hg pressure. Filters are placed in scintillation vials after collection and immediately frozen at -80C. After the cruise is complete, the samples are transferred into the -80C freezer in the laboratory at IOS in room 1310 until analysis. The chlorophyll-a and phaeophyin concentrations in seawater are measured using the Holm-Hansen et al. (1965) fluorometric acidification method. Samples are extracted in 90% (v/v) acetone at -20C for 24 +/- 1 hours in the lab. After extraction at -20C, the samples are warmed to room temperature for 1 hour prior to analysis. The sample are analyzed on a Turner-Trilogy 7200-000 fluorometer. The fluorometer was calibrated with commerically pure chlorophyll-a standard (Sigma) prior to analysis. The current calibration being used can be found in the Raw Data Processing sheet. Fluorescence readings taken before and after acidification with 1.2N HCl were used to calculate chlorophyll and phaeopigment concentrations as per Holm-Hansen et al 1965. Chlorophyll samples were analyzed 6-7 weeks after the cruise. For details see document QF2023-029_CHL*.xlsx. Nutrient samples were collected in plastic tubes and quick frozen in aluminum blocks stored in a -20C freezer. For samples 400m and deeper a second set of samples are collected and stored cool for silicate analysis. All samples were returned to the Institute of Ocean Sciences and they were analyzed using an Astoria-Pacific Analyzer following methods described in IOS Nutrient Methods (1996) Barwell-Clarke and Whitney.
