All samples were returned to the Institute of Ocean Sciences and analyzed 15.5 weeks after the cruise on a Turner Trilogy7200-000 fluorometer, using methods described in Holm-Hansen et al. (1965). Fluorescence readings taken before and after sample acidification were used to calculate chlorophyll and phaeopigment concentrations. For further details on methodolgy, see document QF 2022-015_CHL_*.xlsx Methodology sheet. NOTE: Flags and comments apply to chlorophyll values only. No flags or comments are assigned for Phaeo-Pigment values Oxygen samples were analyzed in the laboratory at the Institute of Ocean Sciences using an automated Winkler titration system (Metrohm Dosimat model 876, (Metrohm Dosimat model 876 and a UV light source and detector with a 365nm filter controlled by LV02_876 software designed and constructed by Scripps Institution of Oceanography) with modifications based on Carpenter (1965) and adhering to WOCE protocols (Culberson 1991). For details including a precision analysis, see document QF2022-015OXY*.xlsx. Salinity samples were collected in 200 mL type ll glass bottles with disposable nylon inserts and screw caps supplied by Ocean Scientific International Limited. They were analyzed in a temperature-controlled lab on a Guildline 8400B Salinometer standardized with IAPSO standard seawater 35 - 43 days after collection. For details including a duplicate analysis, see document QF2022-015SAL*.xlsx. Nutrient samples were collected in plastic tubes and quick frozen in aluminum blocks stored in a -20C freezer. For samples 400m and deeper a second set of samples are collected and stored cool for silicate analysis. All samples were returned to the Insitute of Ocean Sciences and they were analyzed using an Astoria-Pacific Analyzer following methods described in IOS Nutrient Methods (1996) Barwell-Clarke and Whitney. For details including a duplicate analysis, see document QF2022-015nuts*.xlsx. References: 1. Barwell-Clarke, J. and Whitney, F. 1996. Institute of Ocean Sciences Nutrient Methods and Analysis. There were 2 WetLabs CStar transmissometers in use during this cruise: Channel Transmissometer refers to sensor #1185DR (650nm - red) Channel Transmissometer:Green refers to sensor #1883DG (530nm - green) For comparison with other Institute of Ocean Sciences cruises, note that the transmissometer wavelength is 650nm unless otherwise stated. Seapoint Fluorescence values were unbelievably low, likely due to an error in the gain entered in the configuration file. All values were multiplied by 10. After recalibration, the comparison between Seapoint fluorescence and extracted chlorophyll samples exhibited the usual pattern or fluorescence reading higher than extracted chlorophyl when CHL was <2.5ug/L and lower when CHL was >2.5ug/L. While the CTD fluorescence data are expressed in concentration units, it is recommended that users check extracted chlorophyll values where available. Dissolved oxygen data were recalibrated in two sections as a large change was found in comparison with titrated samples between casts #39 and #51. While calibration drift may be a factor in the change, there was a notable change in oceanic conditions between 180m and 450m that likely affected the response of the oxygen sensor. Channel Salinity:T1:C1 was found to be lower than bottles by approximately 0.002psu, but bottle values are likely a little high due to incomplete flushing of Niskin bottles and evaporation or desorption of glass particles into samples. CTD salinity is likely within +/- 0.002psu. No recalibration was applied to salinity. For details on the processing see the report: 2022-015_Processing_Report.doc. --------------------------------------------------------------------------------- Channel Fluorescence was removed because the values found were extremely low and compared very badly with extracted chlorophyll samples.
