Set event to last 4 characters of file name $END *COMMENTS Sample_Number 14: CHL: extracted sample was at room temp for 3 days before analysis; Rb/Ra looks good Analysis methods: ----------------------- Chlorophyll samples were filtered onto 25mm GF/F filters and stored in cryovials in liquid nitrogen dry shipper onboard, then stored at IOS at -80C prior to analysis Samples were extracted in 90% acetone at -20C for 24 hours in the lab and analyzed on a Turner 10AU flourometer calibrated with commercially pure chlorophyll a standard (Sigma) Fluorescence readings taken before and after acidification were used to calculate chlorophyll and phaeopigment concentrations (Holm-Hansen et al 1965). Chlorophyll samples were analyzed at IOS in Room 2423 ~2 weeks after the cruise. The average of two samples is reported. Variability is assessed as the CV% (std dev / mean*100). Flags and comments apply to chlorophyll values only. Chlorophyll values ranged from 2.22 to 19.92ug/l in 16 samples. Average %CV for this cruise was 2.02 with 0 out of 6 duplicate pairs having a CV>10% and 0 duplicate pairs having a CV>30%. Our average dataset %CV is 3.80 % for 2013 - 2018 so the overall quality of this dataset is excellent. For details see worksheet “CV%” in file QF 2019-044 CHL*.xlsx. Salinity samples were collected in 200 mL type II glass bottles with disposable nylon inserts and screw caps supplied by Ocean Scientific International Limited. They were analyzed in a temperature-controlled lab on a Guildline 8400B Salinometer standardized with IAPSO standard seawater within 55-58 days after collection. For details, see document QF 2019-044 SAL*.xlsx. Nutrient samples were collected in plastic tubes and quick frozen in aluminum blocks stored in a -20C freezer. All analysis was performed on frozen samples. They were analyzed using an Astoria analyzer following methods described in IOS Nutrient Methods (1996) Barwell-Clarke, J. and F. Whitney. Oxygen samples were analyzed at sea using an automated Winkler titration system (Metrohm Dosimat model 876, a UV light source and detector with a 365 nm filter controlled by LVO2_876 software designed and constructed by Scripps Institution of Oceanography) with modifications based on Carpenter (1965) and adhering to WOCE protocols (Culberson 1991). The CTD data in these files were extracted from the CTD files run at the same sites, but there is some uncertainty in the depth from which the samples were taken. In the case of surface samples, the CTD data are only available from a little lower in the water column. Conductivity and Fluorescence:URU:Wetlabs data are nominal and unedited except that some records were removed in editing temperature and salinity. Calibration sampling for salinity and dissolved oxygen was done at 3 sites. There is some uncertainty about the depth at which the bottles fired though wire angles were not reported to have been high. Further, there are likely errors due to incomplete flushing of Niskin bottles which also leads to bottles containing water from above the nominal depths. CTD salinity values were higher than the bottle samples by a median of 0.008psu. Some of that difference may be due to the samples coming from higher in the water column where salinity is lower, though errors due to delayed sample analysis may have offset those errors to some extent. No recalibration was applied to the CTD salinity. Similarly, while Oxygen:Dissolved:SBE values appeared to be low, this would be partly due to samples coming from higher in the water column where values are usually higher. Some calibration drift is expected towards lower values, but the error is likely <2%. No recalibration was applied to Oxygen:Dissolved:SBE. Warning: While the CTD fluorescence data are expressed in concentration units, they do not always compare well to extracted chlorophyll samples.
