They were analyzed in a temperature-controlled lab on a Guildline 8400B Salinometer (S/N 68572) standardized with IAPSO standard seawater 5 months after collection. Calculated Standard pool of the mean (Sp) was 0.0034 using 25 out of 26 replicates. The unused replicate was rejected using the Chauvenet criteria. The following comments refer to channels Nitrate_plus_Nitrite, Silicate and Phosphate; these data were analyzed at the Institute of Ocean Sciences. Nutrient samples were collected in polystyrene tubes, quick frozen in aluminum blocks and stored in -20°C freezer until analysis at IOS, about six months later in January 2014. Samples were analyzed on a three-channel Technicon Nutrient Auto Analyser, following methods described in Barwell-Clarke and Whitney (1996). Full details are in files “Nutrients_C3O_2013-05SWL_04.02.2014.docx and “2013-05_C3O_FrozenNutrientData_2016-08-08.xlsx”. There are two sets of nutrient samples. The following comments refer to channels Nitrate_plus_Nitrite2, Silicate2 and Phosphate2 and Ammonium: Nutrient samples (nitrate+nitrite, orthophosphate,silicate and ammonium) were analyzed at Chesapeake Biological Laboratory of the University of Maryland Center for Environmental Science (CBL-UMCES, http://www.umces.edu/nutrient-analytical-services-laboratory ). They were filtered from the Niskin bottles using 0.45 µM nylon syringe filters and frozen at -20°C until analysis at UMCES. Samples were thawed for 2 weeks at 8 degrees C before analysis. using a Technicon Bran & Luebbe AutoAnalyzer II, following these methods: Nitrate+Nitrite: Technicon Ind. Method No.158-71, 1977 and USEPA 1979 Method 353.2. Silicate: Technicon Ind. Method No.811-86T, 1986. Phosphate: Technicon Ind. Method No.155-71W, 1973, 1971 USEPA 1979 Method 365.1 and Froelich, 1978. NH4: References Standard Methods 4500-NH3 G-1997 (for further details, see CBL-UMCES_Ammonium Method 2017-3_3.pdf) Data provided by Lee Cooper (UMCES) Oxygen isotopic analysis water samples were collected in 10 or 20 mL glass scintillation vials, sealed with Polyseal caps and wrapped with Parafilm as a precaution to prevent evaporation and isotopic fractionation. Samples were returned to the Chesapeake Biological Laboratory of the University of Maryland Center for Environmental Science. Water samples were analyzed by equilibration with carbon dioxide using a Thermo Fisher Gas Bench II peripheral linked to a continuous flow Delta V Plus isotope ratio mass spectrometer. The oxygen isotope composition is referenced to Vienna-Standard Mean Ocean Water (V-SMOW): (V-SMOW): d18O = ((Rsample-Rstandard) / Rstandard) *1000 per mille, where R=18O/16O and V-SMOW is Vienna Standard Mean Ocean Water, as distributed by the International Atomic Energy Agency. Analytical precision was better than ±0.1 per mille and was assessed by analysis of in-house water standards during sample analysis and calibration to international water isotope standards (V-SMOW, V-SLAP, GISP). Data corrections were made based upon analysis of the in-house standards relative to international standards, which were normalized as per recommendations of Paul et al. [2007]. Data provided by Lee Cooper (UMCES) DIC/Alkalinity samples were collected in 250 mL glass reagent bottles, preserved with HgCl2, sealed with grease and stored at 4°C, until analysis at IOS about 7 months after collection. Samples were first analysed for DIC on a SOMMA-1 (Single-Operator Multi-Metabolic Analyzer), a computer controlled automated dynamic headspace analyser. The coulometer system is similar to that described by Johnson et al (1993) and was calibrated with Dickson CRM seawater (Batch 133 with a concentration of 2021.12 µmol/kg) (DOE 1994; Dickson 2001; Dickson et al. 2003) supplied by Andrew Dickson (Scripps Institute of Oceanography, San Diego, USA). There was only one duplicate collected so the pooled standard deviation could not be calculated. Samples were then analyzed for alkalinity by means of potentiometric titration using 0.1N HCl/0.6N NaCl, using a program written by Paul Covert, PMEL, University of Washington which is based on Andrew Dickson’s, SCRIPPS system. The mean accuracy (%recovery) was calculated to be 99.87%, ranging from 99.76 to 100.13%. Only one sample was collected in duplicate and so, the pooled standard deviation could not be calculated. Filters were frozen at -20°C initially to fracture cell membranes, then 10 mL of 90% acetone was added and samples stored in the dark for 24 hours at 2°C. Chlorophyll concentration in acetone was then analyzed shipboard on a Turner 10AU fluorometer without acidification (Welschmeyer, 1994). The fluorometer was calibrated with a pure chlorophyll-a standard obtained from Turner Designs, (Turner Designs Part No.10-850) before and after all sampling, with use of a secondary solid standard (Part No.10-AU-904) during sampling to identify any possible instrument drift. There was no calibration sampling for dissolved oxygen, so values are reported with 1 less significant figure than usual. The post-cruise factory calibration indicates salinity drift was insignificant. For further processing details see the processing report 2013-05_Processing_Report.doc. --------------------------------------------------------------------------------------- This research cruise is part of the Distributed Biological Observatory (DBO) effort, which is an international effort to make repeat oceanographic observations at chosen stations in the Bering and Chukchi Seas and in Barrow Canyon. It is also a continuation of the Canada Three Oceans project “C3O” which was part of Canada’s contribution to the International Polar Year (IPY) research effort. Collaborators include University of Maryland, Center for Environmental Science (UMCES), Clark University and NOAA in the US, and the Universities of Victoria and British Columbia in Canada.
