Analysis methods: ----------------- Chlorophyll samples were filtered onto 25mm GF/F filters and stored in glass scintillation vials. Filters were frozen at -20 C initially to fracture cell membranes, then 10 mL of 90% acetone was added and samples stored in the dark for 24 hours at 2C. Chlorophyll concentrations in acetone were then analyzed on shipboard on a Turner 10AU fluorometer without acidification (Welschmeyer, 1994). The fluorometer was calibrated with a pure chlorophyll a standard obtained from Turner Designs, (Turner Designs Part No.10-850) before and after all sampling, with use of a secondary solid standard (Part No.10-AU-904) during sampling to identify any possible instrument drift. Salinity samples were collected in 200 mL type ll glass bottles with disposable plastic inserts and screw caps supplied by Ocean Scientific International Limited. They were analyzed in a temperature-controlled lab on a Guildline 8400B Salinometer (S/N 68572) standardized with IAPSO standard seawater 1 month after collection. Full details including a duplicate analysis are in file "2010 SWL Salt Data.xls". Nutrient samples (nitrate+nitrite, orthophosphate and silicate) were analysed at the University of Maryland Center for Environmental Science (UMCE). Samples were filtered from the Niskin bottles using GF/F and frozen at -20C for analysis at UMCE about 3 months after collection using a Technicon Bran & Luebbe AutoAnalyzer II sampler, following methods listed on http://nasl.cbl.umces.edu/ Nitrate+Nitrite2: Filtered samples are passed through a granulated copper-cadmium column to reduce nitrate to nitrite. The nitrite (originally present plus reduced nitrate) then is determined by diazotizing with sulfanilamide and coupling with N-1- naphthylethylenediamine dihydrochloride to form a colored azo dye. Nitrate concentration is obtained by subtracting the corresponding nitrite value from the nitrite + nitrate concentration. See Technicon Ind. Method No.158-71, 1977 and USEPA 1979 Method 353.2. Silicate2: This reaction is based on the reduction of silicomolybdate in acidic solution to “molybdenum blue” by ascorbic acid. Oxalic acid is added to minimize interference from phosphates. See Technicon Ind. Method No.811-86T,1986. Phosphate2: Filtered samples are mixed with a sulfuric acid-antimony-molybdate solution, and subsequently with an ascorbic acid solution, yielding an intense blue color suitable for photometric measurement. See Technicon Ind. Method No.155-71W, 1973, 1971 USEPA 1979 Method 365.1 and Froelich, 1978. Dissolved inorganic carbon (DIC) was determined using coulometric analysis. Total alkalinity (Alk) was determined by potentiometric titration. Detailed documentation on the DIC and ALK sampling, analysis and precision estimates can be found in file “2010-05-DIC and ALK data.xlsx.” Oxygen isotopic analysis water samples were collected in 10 or 20 mL glass scintillation vials, sealed with Polyseal caps and wrapped with Parafilm as a precaution to prevent evaporation and isotopic fractionation. Samples were returned to the Chesapeake Biological Laboratory of the University of Maryland Center for Environmental Science. Water samples were analyzed by equilibration with carbon dioxide using a Thermo Fisher Gas Bench II peripheral linked to a continuous flow Delta V Plus isotope ratio mass spectrometer. The oxygen isotope composition is referenced to Vienna-Standard Mean Ocean Water (V-SMOW): (V-SMOW): d18O = ((Rsample-Rstandard) / Rstandard) *1000 per mille, where R=18O/16O and V-SMOW is Vienna Standard Mean Ocean Water, as distributed by the International Atomic Energy Agency. Analytical precision was better than ±0.1 per mille and was assessed by analysis of in-house water standards during sample analysis and calibration to international water isotope standards (V-SMOW, V-SLAP, GISP).
