The filters are placed in glass scintillation vials and immediately frozen at -20ēC until analysis. The chlorophyll is extracted in 10 ml of 90% acetone for 24h at -20ēC, then brought to room temperature and the extract is decanted and measured using a Turner Designs 10-AU-005-CE fluorometer, which has been calibrated with solutions made from commercially prepared pure chlorophyll a. Fluorometer readings are taken before and after acidification of the extract with two drops of 1.5N HCl. Average of two samples is reported. Variability is assessed as the % (std dev/mean*100) and samples are flagged if >10%. HPLC samples were filtered onto 47mm GF/F filters and stored at -80C prior to analysis. Samples were extracted in 95% methanol at -20C for 24 hours in the lab and analyzed on a WATERS 2695 HPLC separations system as detailed in Nemcek and Pena, 2014. Analysis was performed ~6 months after collection. The average of two samples is reported. Variability is assessed as the %CV (std dev/mean*100) for duplicate pairs. All pigments below the limit of detection (LOD) are assigned a zero value. TChl-a is the sum of Chl-a, DVchl-a and Chlide-a. For further information see file QF 2009-064_HPLC*.xlsx. Oxygen samples are analyzed on an automated Winkler titration system following the procedures of Carpenter (1965). Samples were analyzed at sea using an automated titration system consisting of a Brinkman Dosimat (model 665) and a PC 950 Colorimeter. Endpoint titers were optimized in post-processing using a best fit linear regression utility. File 2009-64oxygen.xls includes the raw data and duplicate analysis. Salinity samples were collected in glass bottles and analyzed on Guildline Autosal model 8400B, S/N 69086, at IOS. Salinometers are standardized with IAPSO standard seawater. Nutrient samples were collected in plastic tubes and stored frozen and later analysed at IOS using an Astoria analyzer following methods described in Barwell-Clarke and Whitney (1996). Then a sudden shift occurred so that values between September 2009 and July 2010 were very low, ~25%/m. After the shift, drift continued at a similar rate to that before the shift. There is some pressure dependence after the shift, so that transmissivity is decreasing with pressure even in deep offshore waters. In August 2010 a study was made of transmissivity that led to a decision to apply post-processing corrections to all cruises between March 2008 and June 2010. Transmissivity data from this cruise were corrected by multiplying the original values by a correction factor 2.087, which was based on assumptions that deep offshore transmissivity from February 2010 and June 2010 Line P cruises should be about 62%/m and that drift was linear with time. The corrections produced reasonable results for other cruises in late 2009.
