The filters are placed in glass scintillation vials and immediately frozen at -20ēC until analysis. The chlorophyll is extracted in 10 ml of 90% acetone for 24h at -20ēC, then brought to room temperature and the extract is decanted and measured using a Turner Designs 10-AU-005-CE fluorometer, which has been calibrated with solutions made from commercially prepared pure chlorophyll a. Fluorometer readings are taken before and after acidification of the extract with two drops of 1.5N HCl. Average of two samples is reported. Variability is assessed as the % (std dev/mean*100) and samples are flagged if >10%. HPLC samples were filtered onto 25mm GF/F filters and stored at -80C prior to analysis. Samples were extracted in 95% methanol at -20C for 24 hours in the lab and analyzed on a WATERS 2695 HPLC separations system as detailed in Nemcek and Pena, 2014. Analysis was performed ~1 month after collection. The average of two samples is reported. Variability is assessed as the %CV (std dev/mean*100) for duplicate pairs. All pigments below the limit of detection (LOD) are assigned a zero value. TChl-a is the sum of Chl-a, DVchl-a and Chlide-a. For further information see file QF 2009-026_HPLC*.xlsx. Oxygen samples are analyzed on an automated Winkler titration system following the procedures of Carpenter (1965). Samples were analyzed at sea using an automated titration system consisting of a Brinkman Dosimat (model 665) and a PC 900 Colorimeter. Salinity samples were collected in glass bottles and analyzed on a Guildline model 8400B Autosal at IOS. Salinometers are standardized with IAPSO standard seawater. Nutrient samples were collected in plastic tubes and stored frozen and later analysed at IOS using an Astoria analyzer following methods described in Barwell-Clarke and Whitney (1996). Dec. 18, 2009. ********************************************************************************* Transmissometer #1005DR was calibrated in March 2008, and drifted significantly but steadily until July 2009; then a sudden shift occurred, so that maximum values between September 2009 and July 2010 were very low, ~25%/m. In August 2010 a study was made of transmissivity that led to a decision to apply post-processing corrections to all cruises between March 2008 and June 2010. Transmissivity data from this cruise were corrected by multiplying the original values by a correction factor, which was based on assumptions that deep offshore transmissivity from a June 2009 cruise should be about 62%/m and that drift was linear with time between March 2008 and July 2009. The corrections produced reasonable results for all cruises in that time period.
