There is no bottle salinity available, but based on nutrient, CHL and dissolved oxygen samples, and observations during other casts, it is believed that the bottle closed at about the same time as Niskin #8, so the CTD data from that level is likely to be a better match to the bottle contents. Sample #447: ALL PROPERTIES: Niskin bottle #14 closed prematurely on many occasions during this cruise, and it appears to have done so during this cast. There is no bottle salinity available, but based on nutrient, CHL and dissolved oxygen samples, and observations during other casts, it is believed that the bottle closed at about the same time as Niskin #8, so the CTD data from that level is likely to be a better match to the bottle contents. It is believed that the bottle closed at about the same time as Niskin #9, so the CTD data from that level is likely to be a better match to the bottle contents. Analysis methods: ----------------- Chlorophyll samples were analyzed at sea using a Chemistry Turner Designs fluorometer which was calibrated in May 2007 using pure chlorophyll a. Chlorophyll precision was determined by analyzing replicate samples drawn from one Niskin. For details see worksheet “Precision” in file QF 2008-01CHL.xls. Oxygen samples are analyzed on an automated Winkler titration system following the procedures of Carpenter (1965). Samples were analyzed at sea using an automated titration system consisting of a Brinkman Dosimat (model 665) and a PC 900 Colorimeter. For details on precision see file 2008-01-DO-duplicates.xls. Salinity samples were collected in glass bottles and analyzed on a Guildline model 8400B Autosal or a Guildline Portasal 8410A. Salinometers are standardized with IAPSO standard seawater. See file 2008-01-duplicates-sal.xls for information on precision. Nutrient samples were collected in plastic tubes and analyzed fresh at sea using a Technicon AAII autoanalyzer following methods described in Barwell-Clarke and Whitney (1996). For details on nutrient duplicates and precision see file QF 2008-01nuts.xls. References: 1. Carpenter, J.H. 1965. The Chesapeake Bay Institute Technique for the Winkler Dissolved Oxygen Method. Canadian Technical Report of Hydrography and Ocean Sciences, No. 182, 43 pp. WARNING: All samples from Niskin Bottles #13 to #18 should be treated with care. While the CTD data corresponds to the level at which the bottle was fired, in many cases the bottle closed earlier. Notes in the headers of individual files indicate the probable level at which bottles closed, where evidence exists to make that estimate. Chlorophyll precision was determined by analyzing replicate samples drawn from one Niskin. For details see worksheet “Precision” in file QF 2008-01CHL.xls. HPLC samples were filtered onto 25mm GF/F filters and stored at -80C prior to analysis. Samples were extracted in 95% methanol at -20C for 24 hours in the lab and analyzed on a WATERS 2695 HPLC separations system as detailed in Nemcek and Pena, 2014. Analysis was performed ~2 weeks after collection. The average of two samples is reported. Variability is assessed as the %CV (std dev/mean*100) for duplicate pairs. All pigments below the limit of detection (LOD) are assigned a zero value. TChl-a is the sum of Chl-a, DVchl-a and Chlide-a For further information see file QF 2008-001_HPLC*.xlsx. While the CTD data corresponds to the level at which the bottle was fired, in many cases the bottle closed earlier. Notes in the headers of individual files indicate the probable level at which bottles closed, where evidence exists to make that estimate.
