They were analyzed at sea on a Guildline 8400B Guildline Salinometer (S/N 69086) standardized with IAPSO standard seawater within 9 - 22 days after collection. For details including a duplicate analysis, see document "Salts 2007-19 for merging*.xls". Oxygen samples were collected in glass flasks and analyzed on board using an automated Micro-Winkler titration system following the procedures of Carpenter (1965). The titration was performed with a Metrohn Dosimat 665 and the end point was detected using a Brinkmann probe Colorimeter PC910 (SN: 910-358). Software written at the Institute of Ocean Sciences, NewAutoOxy.exe, was used to calculate dissolved oxygen (ml/L). Post-cruise corrections included changing the volume dispensed for the potassium iodate standard and flask volumes following re-calibrations in 2008 and 2009. Oxygen values changed from -0.04 to +0.06 ml/l. Note that Casts 1 through 10 are flagged bad as there was a problem with the colorimeter probe. The probe was changed out after Cast 10 however there was another unknown problem that compromised casts 17, 18 and 22 which have also been flagged bad. Nutrient samples (nitrate plus nitrite, silicate and orthophosphate) were collected in polystyrene test tubes and quick frozen in a -20 degree C freezer. Samples were analyzed at the Institute of Ocean Sciences in March 2008, using a Technicon auto-analyzer following methods described in Barwell-Clarke and Whitney (1996). For more details see document “Nutr_Frozen_2007-19_14Nov2008.xls Ammonium samples were collected and analysed onshore at the University of Victoria. The ammonium method used is a simplification of the fluorometric technique of Holmes et al, 1999 The minimum detectable level for NH4 is 0.05 nmol/L, so “0” values should be interpreted as <0.05nmol/L. For full data set see document 2007-19_LSSL_Chem_RAW_*.xls. Dissolved Inorganic Carbon (DIC) and Alkalinity: DIC and Alkalinity were collected in 250 or 500 glass bottles. Samples were kept at 4 degree C until analysis and preserved with HgCl2. Alkalinity ("Alkalinity:Total") and DIC ("Carbon:Dissolved:Inorganic") were measured from the same sample bottle at select stations. In addition, more frequent samples of Alkalinity were collected at most stations and have been labelled "Alkalinity:Total:Potentiometric" to distinguish this set of separate samples, even though both sets of Alkalinity samples were measured using potentiometric titration. DIC was analyzed at the Institute of Ocean Sciences using a SOMMA (Single-Operator Multi-Metabolic Analyzer) - Coulometer system to determine DIC. Alkalinity samples were analyzed using an automated potentiometric titration system to determine the total alkalinity. Alkalinity samples paired with DIC and those sampled into separa bottles were both analysed at the Institute of Ocean Sciences. Analysis was completed by May 200 Total Chlorophyll-a (>0.7um) samples were collected into 1L brown Nalgene sample bottles. Sampl size ranged from 250 to 900ml and was filtered onto 25 mm glass fiber filters (Whatman GF75, >0. Fraction chlorophyll samples were filtered through a 3 µm filter. Both were filtered under 5 ps vacuum, placed into glass scintillation vials and either extracted immediately or frozen at -20 for a few weeks until they were analyzed onboard ship. For analysis, 10 ml of 90% acetone was a to the scintillation vial which was then put into a -20 degree C freezer for 24 hours. The samples were allowed to adjust to room temperature for an hour before being read with the fluorometer. Chlorophyll-a and phaeo-pigment levels were measured with a Turner Designs 10 AU-00 Field Fluorometer SN:5152FRXX, calibrated with commercially pure chlorophyll a standard (Sigma). The sample was acidified with 2 drops of 1N hydrochloric acid to obtain the phaeo-pigment readin (Holm-Hansen et al 1965). Flags and comments apply to chlorophyll values only.
