Analysis methods: ----------------- Salinity samples are collected in glass bottles and analyzed on Guildline Salinometers which are standardized with IAPSO standard seawater. For this cast two Guildline Portasal Salinometers were used. Oxygen samples are analyzed on an automated Winkler titration system following the procedures of Carpenter (1965). A Brinkmann model 665 Dosimat and model PC910 Colorimeter is controlled by Visual-Basic program to titrate the oxygen samples. Samples were analysed fresh at sea using the new Astoria system following methods described in Barwell-Clarke and Whitney (1996). Chlorophyll samples are collected in calibrated polycarbonate bottles (~330ml), then filtered onto 25mm MFS GF/75 filters at <70mmHg vacuum. The filters are placed in glass scintillation vials and immediately frozen at -20ºC until analysis. The chlorophyll is extracted in 10 ml of 90% acetone for 24h at -20ºC, then brought to room temperature and the extract is decanted and measured using a Turner Designs Model 10 fluorometer, which has been calibrated with solutions of commercially prepared chlorophyll. Chlorophyll samples were analyzed on board ship. HPLC samples were filtered onto 25mm GF/F filters and stored at -80C prior to analysis. Samples were extracted in 95% methanol at -20C for 24 hours in the lab and analyzed on a WATERS 2695 HPLC separations system as detailed in Nemcek and Pena, 2014. Analysis was performed ~6-24 months after collection. The average of two samples is reported. Variability is assessed as the %CV (std dev/mean*100) for duplicate pairs. All pigments below the limit of detection (LOD) are assigned a zero value. TChl-a is the sum of Chl-a, DVchl-a and Chlide-a For further information see file QF 2007-013_HPLC*.xlsx.
