Sampling Mathod --------------- Water from Saanich Inlet was sampled using a Masterflex Persistatic pump set at low speed using clear Tygon tubing, except at the pump head, where clean silicone tubing was used. The tubing was attached to a weighted wire which had been marked at 1m intervals. The tubing and wire were lowered to the desired depth, the pump was run until the tubing contained water from the desired depth, then samples were taken from the outlet. Analysis methods: ----------------- August 7-21 nutrient samples, analyzed on August 21, 2003 by M. Quenneville August 22, 25, 26 nutrient samples frozen and analyzed onboard cruise 2003-2 by Janet Barwell-Clarke. Nutrient samples are analyzed frozen using a Technicon AAII autoanalyzer following methods described in Barwell-Clarke and Whitney (1996). The extracted chlorophyll was analyzed by Melanie Quenneville. Sample numbers were not assigned. Chlorophyll samples are collected in duplicate in calibrated polycarbonate bottles (~330ml), then filtered onto 25mm MFS GF/75 filters at <70mmHg vacuum. The filters are placed in glass scintillation vials and immediately frozen at -20ºC until analysis. The chlorophyll is extracted in 10 ml of 90% acetone for 24h at -20ºC, then brought to room temperature and the extract is decanted and measured using a Turner Designs 10-AU-005-CE fluorometer, which has been calibrated with solutions of commercially prepared chlorophyll.
