Analysis: Sept. 16 1997 by Linda White Analysis methods: ----------------- Salinity samples are collected in glass bottles and analyzed on Guildline model 8410 Portasal Salinometers which are standardized with IAPSO standard seawater. Nutrient samples are analyzed frozen using a Technicon AAII autoanalyzer following methods described in Barwell-Clarke and Whitney (1996). Chlorophyll samples are collected in duplicate in calibrated polycarbonate bottles (~330ml), then filtered onto 25mm MFS GF/75 filters at <70mmHg vacuum. The filters are placed in glass scintillation vials and immediately frozen at -20ºC until analysis. The chlorophyll is extracted in 10 ml of 90% acetone for 24h at -20ºC, then brought to room temperature and the extract is decanted and measured using a Turner Designs 10-AU-005-CE fluorometer, which has been calibrated with solutions of commercially prepared chlorophyll. Canadian Technical Report of Hydrography and Ocean Sciences, No. 182, 43 pp. ___________________________________________ NOTES FROM LINDA WHITE ON NUTRIENT ANALYSIS ------------------------------------------- Phosphate: for salinities less than 20: correction= .040-((.0019*sal)+.0012) the correction factors should be added to the measured nutrient value. Nitrate/silicate samples were defrosted for about 20 h at room temperature. Phosphate samples were defrosted for about 1 h in moving air. Samples from stations 43-63 were stored in longer test tubes; combined with a low sample volume made analysis transferred impossible before transferring into more appropriate test tubes. This transfer is susceptible to contamination and error. Silicate at stations 29, 30, 43, 56, 58, and 60 was higher at the surface than at 10 m. This may indicate the presence of silt or other terrestrial input in the water. Phosphate and silicate levels at stations 13 and 16, 198m were higher than the expected levels (phosphate around 4, silicate above 100uM) The method described by Parsons, Maita, and Lalli for Total Dissolved Organic Nitrogen was followed, except that the samples were processed in a pressure cooker after addition of the persulfate reagent, and the sample was not buffered after addition of the acid. The samples sat for 3 days after pressure cooking before the acid was added to dissolve the precipitate. They were in their original Teflon vessels. I would recommend that a larger volume of water be sampled if these samples were collected again. A 40 ml sample is required for the digestion, and about 20 ml extra is used for rinsing the pipettor. The DON data are of very poor quality, due to either sampling/storage problems or analytical error.The methodology for this technique is well established, except that the samples are usually acidified and analyzed soon after autoclaving. The samples were stored frozen in PVC containers. There was a moderate quantity of seawater in the bags after the samples had been removed; it is likely the containers were overfilled and some sample extruded from the containers. This could account for some of the variability, particularly of the replicates. Of the DON data which appear in the normal range (4-6 for offshore, W. Richardson, p. comm), there do not appear to be values in great excess of oceanic levels. ******************************************************************************* The bottle depths are approximate.
