Sample Number 336: Bottle Number 14 - top valve slightly open; SAL:Bottle - outlier in COMPARE. Analysis methods: ----------------- Salinity samples were collected in glass bottles and analyzed on Guildline model 8400B Autosal Salinometers which are standardized with IAPSO standard seawater. Oxygen samples were analyzed on an automated Winkler titration system following the procedures of Carpenter (1965). A Brinkmann model 665 Dosimat and model PC910 Colorimeter is controlled by Visual-Basic program to titrate the oxygen samples. Nutrient samples were analyzed fresh at sea using a Technicon AAII autoanalyzer following methods described in Barwell-Clarke and Whitney (1996). For details on duplicate samples and precision see file 2007-001_NUT.pdf. Chlorophyll samples were collected in calibrated polycarbonate bottles (~330ml), then filtered onto 25mm MFS GF/75 filters at <70mmHg vacuum. The filters were placed in glass scintillation vials and immediately frozen at -20ºC until analysis. The chlorophyll was extracted in 10 ml of 90% acetone for 24h at -20ºC, then brought to room temperature and the extract was decanted and measured using a Turner Designs Model 10 fluorometer, which had been calibrated with solutions of commercially prepared chlorophyll. Chlorophyll samples were analyzed on board ship. The minimum detectable level for DMS is 0.10 nM, so “0” values should be interpreted as <0.10. For details on the collection and analysis of the DMS data see file 2007-001_DMS_report.doc. For details on duplicates see file 2007-001_DMS.pdf. Dissolved inorganic carbon (DIC) was determined using coulometric analysis. Total alkalinity (Alk) was determined by potentiometric titration. For duplicates and precision of DIC and ALK samples see files 2007-001_DIC.pdf and 2007-001_ALK.pdf respectively. HPLC samples were filtered onto 25mm GF/F filters and stored at -80C prior to analysis. Samples were extracted in 95% methanol at -20C for 24 hours and analyzed on a WATERS 2695 HPLC separations system as detailed in Nemcek and Pena, 2014. Analysis was performed ~6-24 months after collection. The average of two samples is reported. Variability is assessed as the %CV (std dev/mean*100) for duplicate pairs. All pigments below the limit of detection (LOD) are assigned a zero value. TChl-a is the sum of Chl-a, DVchl-a and Chlide-a. References: 1. Carpenter, J.H. 1965.
